ABSTRACT

It is unclear how VvLaeA functions in regulating the growth and development of Volvariella volvacea (Bull. ex. Fr.) Sing.. Firstly, bioinformatics analysis of VvLaeA was carried out in this study. Subsequently, the Vvgpd promoter and the open reading frame (ORF) fragment of VvlaeA were amplified and fused by polymerase chain reaction (PCR). The fusion fragment was cloned into the pK2 (bar) plasmid. The recombinant construct pK2(bar)-OEVvlaeA was transfected into Beauveria bassiana by Agrobacterium tumefaciens-mediated transformation. Finally, the growth and development of the transformants were examined. The results showed that VvLaeA shared a low homology with similar proteins in other fungi. Compared with the wild type, the colony diameter of the transformant was significantly increased. However, the pigment deposition, conidial yields and germination rates were significantly decreased. The overexpression strains were more sensitive to stresses than that of the wild type. Further studies showed the conidial cell wall properties of the transformants were altered, and the expressions of genes related to the conidial development were significantly down-regulated. Collectively, VvLaeA increased the growth rate of B. bassiana strains and negatively regulated the pigmentation and conidial development, which shed a light for the functional identification of straw mushroom genes.

ABSTRACT

Aedes aegypti is a culicide that has gained relevance over the years due to its ability to transmit various viruses that cause diseases in humans that all the years cause high mortality rates in the world population. The main problem is that Ae. aegypti has managed to establish and maintain a close relationship with humans and their habitat, which is why the search for alternatives to control vector populations becomes imperative. The objective of the present work was to study the effects of two Beauveria bassiana strains on Aedes aegypti. Third instar larvae of Ae. aegypti in 250 mL plastic containers were inoculated with the GHA and NB3 strains at different concentrations (1.5 × 104, 1.5× 105, 1.5 × 106 and 1.5 × 107 conidia/mL). The NB3 strain presented highest mortality values with 63% in the highest concentration i.e., 1.5 × 107, while for the GHA strain the highest mortality value was 30.7% at the same concentration. The results showed significant difference in mortality with respect to the strain and days post treatment (P = 0.0001), but not with respect to the conidial concentration (P = 0.634). The average mortality of larvae per day for the NB3 for different concentrations ranged from 20 to 25 larvae per day, while for the GHA daily mortality ranged from 5 to 12 larvae. In post-treatment mortality, the highest mortality was recorded in the third stage larvae for the NB3, while for GHA the highest percentage mortality was observed in individuals who managed to reach the adult state. The findings of the current research depicted the noteworthy role of B. bassiana for the management of an important vector of human disease.


O Aedes aegypti é um culicida que vem ganhando relevância ao longo dos anos devido à sua capacidade de transmitir diversos vírus causadores de doenças em humanos que ao longo dos anos ocasionam altas taxas de mortalidade na população mundial. O principal problema é que Ae. aegypti tem conseguido estabelecer e manter uma relação próxima com o homem e seu habitat, por isso a busca por alternativas para o controle das populações de vetores torna-se imperativa. O objetivo do presente trabalho foi estudar os efeitos de duas cepas de Beauveria bassiana sobre Ae. aegypti. Larvas de terceiro instar de Ae. aegypti em recipientes plásticos de 250 mL foram inoculados com as cepas GHA e NB3 em diferentes concentrações (1.5 × 104, 1.5 × 105, 1.5 × 106 e 1.5 × 107 conídios/mL). A cepa NB3 apresentou os maiores valores de mortalidade com 63% na concentração mais alta, ou seja, 1.5 × 107, enquanto para a cepa GHA o maior valor de mortalidade foi 30.7% na mesma concentração. Os resultados mostraram diferença significativa na mortalidade com relação à cepa e dias pós-tratamento (P = 0.0001), mas não com relação à concentração de conídios (P = 0.634). A mortalidade média de larvas por dia para o NB3 para diferentes concentrações variou de 20 a 25 larvas por dia, enquanto para o GHA a mortalidade diária variou de 5 a 12 larvas. Na mortalidade pós-tratamento, a maior mortalidade foi registrada nas larvas de terceiro estágio para o NB3, enquanto para o GHA o maior percentual de mortalidade foi observado em indivíduos que conseguiram atingir o estado adulto. Os resultados da pesquisa atual retratam o papel notável de B. bassiana no manejo de um importante vetor de doenças humanas.

Entomopathogenic fungi (EPF) now a possible safer microbial control measure that could be considered as a substitute for chemical control of insect pests. Three EPF viz., Metarihizium anisopliae, Isaria furnosoroseus and Beauveria bassiana were evaluated for their virulence against the grubs of Khapra beetle, Trogoderma granarium (Everts) under laboratory conditions. The isolates were applied by two methods viz., diet incorporation and an immersion method with 3rd instar 20 grubs of T. granarium for each. The virulence of EPF was determined using percent mortality. Significantly higher mortality was observed in M. anisopliae applied through immersion (98.33%) and diet incorporation (93.33%) methods followed by B. bassiana (90.83 and 85.83%, respectively). The mortality caused by I. furnosoroseus was statistically lower in immersion and diet incorporation methods i.e. 81.67 and 73.33%, respectively. Based on the immersion method, all EPF were studied for multiple conidial concentration i.e., 1×104, 1×105, 1×106, 1×107 and 1×108 under the same in-vitro conditions. All the isolates were pathogenic to grub of T. granarium at the highest conidial concentration. M. anisopliae was proved the most effective virulent resulting in 98.33% mortality of the pest with LT50 4.61 days at 1 × 108 conidial concentration followed by 90.83 and 81.67 percent mortality with 5.07 and 8.01 days LT50, in the application of B. bassiana and I. furnosoroseus, respectively. M. anisopliae showed higher efficacy and could be considered as promising EPF for the development of myco-insecticides against effective biocontrol of T. granarium.


Os fungos entomopatogênicos (FPE) são agora a possível medida de controle microbiano mais segura, que pode ser considerada um substituto para o controle químico de pragas de insetos. Três EPF viz., Metarihizium anisopliae, Isaria furnosoroseus e Beauveria bassiana foram avaliados quanto à sua virulência contra as larvas do besouro Khapra, Trogoderma granarium (Everts) em condições de laboratório. Os isolados foram aplicados por dois métodos, a saber: incorporação de dieta e um método de imersão com 20 larvas de T. granarium de 3º ínstar para cada um. A virulência do EPF foi determinada usando a mortalidade percentual. Mortalidade significativamente maior foi observada em M. anisopliae aplicado pelos métodos de imersão (98,33%) e incorporação de dieta (93,33%), seguido por B. bassiana (90,83% e 85,83%, respectivamente). A mortalidade causada por I. furnosoroseus foi estatisticamente menor nos métodos de imersão e incorporação de dieta, ou seja, 81,67% e 73,33%, respectivamente. Com base no método de imersão, todos os EPFs foram estudados para múltiplas concentrações de conídios, ou seja, 1 × 104, 1 × 105, 1 × 106, 1 × 107 e 1 × 108 nas mesmas condições in vitro. Todos os isolados foram patogênicos à larva de T. granarium na maior concentração de conídios. M. anisopliae provou ser o virulento mais eficaz, resultando em 98,33% de mortalidade da praga com LT50 4,61 dias na concentração de 1 × 108 conídios seguido por 90,83% e 81,67% de mortalidade com 5,07 e 8,01 dias LT50, na aplicação de B. bassiana e I. furnosoroseus, respectivamente. M. anisopliae apresentou maior eficácia e pode ser considerada como um PFE promissor para o desenvolvimento de micoinseticidas contra o biocontrole efetivo de T. granarium.

ABSTRACT

Coffee is the most appreciated beverage in worldwide; Brazil is the largest producer and exporter of this commodity. Organochlorine endosulfan was banned from the country in 2013 due to its teratogenic agent-related features. Since then, coffee plantations have experienced increased Hypothenemus hampei infestation rates. The aim of the current study is to assess variations in the rates of Coffea arabica fruits brocaded by H. hampei after the application of entomopathogenic fungal species Beauveria bassiana IBCB66. Experiments were carried out with 'Catuaí' and 'Mundo Novo' cultivars between 2018 and 2020, during the borer transit period. Three experiments were carried out based on the application of the aforementioned fungal species on the investigated coffee plant species, both by spraying and sprinkling, at 30-day intervals; 10 fruits were collected per face of each useful plant in each repetition. The experiment has followed a randomized blocks design with five treatments, including the control, and five repetitions, each. Beauveria bassiana Ecobass (IBCB66) wettable powder spray, at the concentration of 2 × 1013·ha­1, was used in experiments I and II. On the other hand, the mix used in experiment III was prepared with blastospores at concentration of 5 × 1012·ha­1 blastospores + 0.1% Silwet. The sprinkling process in all three experiments has used dry aerial conidia at concentration of 2 × 1013·ha­1. Collected data were subjected to analysis of variance (ANOVA), which was followed by Fisher's test at 5% probability level, in the SISVAR software. More than 35,000 fruits were assessed. In addition to variations between experiments, results have evidenced that the rate of brocaded fruits remained high.

Abstract The mortality of the Alabama argillacea and Chrysodeixis includens (Lepidoptera: Noctuidae) larvae caused by the kaolin inert powder and the entomopathogenic fungus Beauveria bassiana were determined under laboratory conditions. Using the caterpillar submersion method, the CG 138 B. bassiana isolate was more pathogenic to A. argillacea than the CG 70, GC 82, ESALQ 634, and ESALQ 645. All five tested isolates caused similar mortality of C. includens. The mortality of first-instar larvae of A. argillacea and C. includens by feeding on leaf-disc impregnated with B. bassiana (CG 138) and kaolin was also determined. Higher A. argillacea mortalities were observed in the B. bassiana (CG 138) treatments, regardless of the presence of kaolin. However, the activity of kaolin + B. bassiana (CG 138) against C. includens was higher than each ingredient alone, indicating an additive action against C. includes larvae. The mortality of A. argillacea and C. includens larvae treated with kaolin + B. bassiana (CG 138) was similar, and the A. argillacea mortality was higher than that of C. includens with kaolin and B. bassiana (GC 138) separated. The treatment kaolin + B. bassiana (CG 138) is promising for the simultaneous management of these two defoliator pests, mainly A. includes. In addition, the monophagous A. argillacea is more susceptible to both kaolin and B. bassiana (GC 138) than the polyphagous C. includens, suggesting that the nutritional ecology plays an important role in the susceptibility of these defoliator species to alternative insecticides.


Resumo A mortalidade de larvas de Alabama argillacea e Chrysodeixis includens (Lepidoptera: Noctuidae), causada pelo pó inerte do caulim e pelo fungo entomopatogênico Beauveria bassiana, foi determinada em condições de laboratório. Usando o método de submersão da lagarta, o isolado CG 138 de B. bassiana foi mais patogênico para A. argillacea que os CG 70, GC 82, ESALQ 634 e ESALQ 645 desse fungo. Os cinco isolados testados causaram mortalidade semelhante de C. includens. A mortalidade de larvas de primeiro ínstar de A. argillacea e C. includens pelo método de alimentação em discos foliares impregnados com B. bassiana (CG 138) e caulim foi, também, determinada. A mortalidade de A. argillacea foi maior nos tratamentos com B. bassiana (CG 138), independentemente da presença do caulim. A atividade do caulim + B. bassiana (CG 138) contra C. includens foi maior que cada ingrediente isolado, indicando ação aditiva contra larvas desse Lepidoptera. A mortalidade de larvas de A. argillacea e C. includens, com caulim + B. bassiana (CG 138), foi semelhante e as de A. argillacea maiores que as de C. includens com caulim e B. bassiana (GC 138) isoladamente. O tratamento caulin + B. bassiana (CG 138) é promissor para o manejo simultâneo dessas duas pragas desfolhadoras, principalmente A. includens. Além disso, a monófaga, A. argilacea, é mais suscetível ao caulim e à B. bassiana (GC 138) que a polífaga, C. includens, sugerindo que a ecologia nutricional desempenha papel importante na suscetibilidade dessas espécies desfolhadoras a inseticidas alternativos.

Abstract Beauveria bassiana is a promising fungus for the biological control of insect pests. The growing costs of conidia production have raised the need to ascertain the efficiency of some low cost substrates. The aim of this study was to analyze the potential use of different raw substrates without nutritional supplement for B. bassiana conidiogenesis. Growth and sporulation were evaluated using 30 g of substrate and 0.3 μL of a conidia suspension (1 x 106 conidia/mL). After 10 days of incubation (70 ± 10% humidity and temperature (T) = 29 ± 1 °C), rice (2.00 x 106 conidia/g substrate), algaroba (2.36 x 106 conidia/g), malt A (1.22 x 106 conidia/g) and malt B (1.75 x 106 conidia/g) showed the highest levels of conidia production. The resulting conidia showed insecticidal activity higher than 80% on coconut termites. These new raw substrates may represent viable alternatives for the production of entomopathogenic fungi for use in the biological control of various insect pests.


Resumo Beauveria bassiana é um fungo promissor no controle biológico de insetos-praga. As crescentes despesas na produção de conídios levantam a necessidade de averiguar a eficiência de alguns substratos de baixo custo. O objetivo deste trabalho foi analisar o potencial de utilização de diferentes substratos brutos para a conidiogênese de B. bassiana. O crescimento e esporulação foram realizados utilizando 30 g do substrato e 0,3 µL da suspensão de conídios (1 x 106 conídios/mL). Após 10 dias de incubação (umidade 70 ± 10% e temperatura T = 29 ± 1° C), o arroz (2,00 x 106 conídios/g de substrato), algaroba (2,36 x 106 conídios/g), malte A (1,22 x 106 conídios/g) e B (1,75 x 106 conídios/g), apresentaram maior produção de conídios. Os conídios produzidos mostraram atividade inseticida sobre o cupim do coqueiro acima de 80% de mortalidade. Estes novos substratos brutos podem representar uma alternativa viável para produção de fungos entomopatogênicos para uso no controle biológico de vários insetos praga.

ABSTRACT

The present study had the objective of evaluating the longevity of A. mellifera workers fed on a diet incorporating commercial entomopathogens, Beauveria bassiana, and Bacillus thuringiensis. It also aimed at verifying possible morphological alterations in the midgut. To this purpose, the entomopathogens used were B. bassiana (Product A) (5.0 × 1011 viable conidia.kg-1), B. thuringiensis (Product B) (2.5 × 109 viable spores.g-1), and B. thuringiensis (Product C) (1.0 × 109 viable spores.g-1); and two controls: T1: sterilized distilled water, and T2: sterilized distilled water + Tween 80® (0.01%). For the bioassays, 2 mL of each treatment were incorporated into Candy paste. For each treatment, 80 bees were individually in flat bottom glass tubes (2.5 cm Ø) covered with voile, containing a piece of cotton soaked in water and Candy paste. These tubes were stored in a B.O.D (30 ± 2°C, R.H 70% ± 10%, 12 h), and mortality was evaluated every six hours, for 10 days. Soon after verifying mortality, two bees per treatment were selected for the removal of their midgut. Midgut samples were processed using standard methodology for Scanning Electron Microscopy (SEM). It was verified that products A, B, and C reduced the longevity of bees when compared to T1 and T2 controls. In the qualitative analyses carried out using SEM, it was not possible to observe external or internal morphological alterations to midgut tissues. Although products A, B, and C cause a reduction in longevity, their presence was not verified when tissues were analyzed using SEM.(AU)


No presente trabalho objetivou-se avaliar a longevidade de operárias de A. mellifera alimentadas com dieta incorporada com os entomopatógenos comerciais Beauveria bassiana e Bacillus thuringiensis, e verificar possíveis alterações morfológicas em seu mesêntero. Para isso, os entomopatógenos utilizados foram B. bassiana (Produto A) (5,0 × 1011 conídios viáveis.kg-1), B. thuringiensis (Produto B) (2,5 × 109 esporos viáveis.g-1), B. thuringiensis (Produto C) (1,0 × 109 esporos viáveis.g-1); e dois controles: T1: água destilada esterilizada e T2: água destilada esterilizada + Tween 80® (0,01%). Para os bioensaios, 2 mL de cada tratamento foram incorporados à pasta Cândi. Para cada tratamento, 80 abelhas foram acondicionadas, individualmente, em tubos de vidro de fundo chato (2,5 cm Ø), cobertos com voile, contendo um pedaço de algodão embebido em água e pasta Cândi. Os tubos contendo as abelhas foram acondicionados em B.O.D (30 ± 2°C, U.R. 70% ± 10%, 12 h), e a mortalidade foi avaliada a cada seis horas, durante 10 dias. Logo após a verificação da mortalidade, foram separadas duas abelhas por tratamento para a retirada do mesêntero. Essas amostras foram processadas em metodologia padrão para Microscopia Eletrônica de Varredura (MEV). Verificou-se que os produtos A, B e C reduziram a longevidade das abelhas quando comparados aos controles T1 e T2. Nas análises qualitativas realizadas com MEV, não foi possível observar alterações morfológicas externas ou internas nos tecidos do mesêntero. Apesar dos produtos A, B e C causarem redução na longevidade, sua presença não foi verificada quando os tecidos foram analisados por MEV.(AU)

ABSTRACT

The use of acaricides is the main control method for Tetranychus ludeni (Acari: Tetranychidae) in horticultural crops. This mite has been recorded causing damage to sweet potato (Ipomoea batatas L.). The use of pathogenic fungi is an alternative to chemical control. The objective was to evaluate the pathogenicity of the fungi Metarhizium anisopliae (Meetch) Sorok, and Beauveria bassiana (Bals.) Vuill. to phytophagous mite T. ludeni in sweet potato plants. Conidial suspensions of M. anisopliae and B. bassiana, at concentrations of 106 and 107 con.mL-1 were applied to sweet potato leaves. After 24 hours, five females of T. ludeni newly emerged were released on the leaves. The bioassay was arranged in a completely randomized design with factorial arrangement 2 × 2 (two species of fungi and two concentrations) plus the control (distilled water), with 10 repetitions per treatment. The evaluation consisted of observing of three biological parameters of the mite: mortality, oviposition, and repellency, after 24, 48, 72, and 96 hours of contact with the fungi. The isolates of M. anisopliae cause high mortality rates of T. ludeni in laboratory. Beauveria bassiana has the potential to suppress future generations of mite, reducing its oviposition rate. Repellency behavior was not observed.(AU)


O uso de acaricidas é o principal método de controle de Tetranychus ludeni (Acari: Tetranychidae) em cultivos hortícolas. Esse ácaro foi registrado causando danos em batata-doce (Ipomoea batatas (L.). A utilização de fungos patogênicos é uma alternativa ao controle químico. O objetivo foi avaliar a patogenicidade dos fungos Metarhizium anisopliae (Meetch) Sorok. e Beauveria bassiana (Bals.) Vuill. ao ácaro fitófago T. ludeni em batata-doce. Suspensões conidiais de M. anisopliae e B. bassiana, nas concentrações de 106 e 107 con.mL-1, foram aplicadas sobre folhas de batata-doce. Após 24 horas, cinco fêmeas recém-emergidas foram liberadas sobre as folhas. O bioensaio foi inteiramente randomizado, com arranjo fatorial 2 × 2 (duas espécies de fungos e duas concentrações) e controle (água destilada), com 10 replicações por tratamento. A avaliação consistiu na observação de três parâmetros biológicos do ácaro: mortalidade, oviposição e repelência, após 24, 48, 72 e 96 horas de contato com os fungos. Os isolados de M. anisopliae causam altas taxas de mortalidade de T. ludeni em laboratório. Beauveria bassiana tem potencial para suprimir futuras gerações do ácaro, reduzindo a taxa de oviposição. Comportamento de repelência nos ácaros não foi observado.(AU)

Se evaluó la efectividad de Baubassil® (Beauveria bassiana) sobre la garrapata Rhipicephalus (Boophilus) microplus. En la fase in vitro, se determinaron la temperatura óptima de germinación del hongo mediante microcultivos a 25, 30 y 35° C, y la mortalidad de las garrapatas. En campo, en un diseño aleatorio, se utilizaron 30 vacas cebuinas; 15 vacas se trataron con Baubassil® una vez por semana con 1 x 10(6) conidias/ml y un grupo control se dejó sin tratamiento. Se observó que a temperaturas de 25, 30 y 35 °C, la germinación del entomopató-geno fue del 97,5, el 94,7 y el 6,3%, respectivamente. En los animales tratados con Baubassil® se obtuvo una efectividad corregida del 96,8%. En el Caribe Seco, Baubassil® constituiría una alternativa de control del ectoparásito, pues no existen registros anteriores en La Guajira del control sostenible de esta garrapata.


The effectiveness of Baubassil® (Beauveria bassiana) on the tick Rhipicephalus (Boophilus) microplus was evaluated. In the in vitro phase, the optimal germination temperature of the fungus was determined by microcultures at 25, 30 and 35 °C and the mortality of ticks. In the field, in a randomized design, 30 cebuine cows; 15 cows were treated with Baubassil® once a week at a 1 x 10(6) conidia/ml, and a control group was left untreated. It was observed that at temperatures of 25, 30 and 35 °C, germination of the entomopathogen was 97.5, 94.7 and 6.3%, respectively. In animals treated with Baubassil® obtained a corrected effectiveness of 96.8% on ticks in the fifth week of application. The use of Baubassil® would be a valid alternative for controlling the cattle tick under the agroecological conditions of the Dry Caribbean.

Rachiplusia nu (Guenée) (Lepidoptera: Noctuidae) is one of the major lepidopteran pests defoliating soybeans (Glycine max Merrill) in Argentina. The combined use of chemical insecticides and entomopathogenic fungi is a promising pest-control option to minimize adverse chemical effects. In this work, we evaluated the interactions between five insecticides-two being considered biorational-and five fungal entomopathogenic strains under laboratory conditions in order to determine the possible usefulness of combinations of these agents against R. nu. The insecticides were tested for compatibility at four doses by in vitro bioassay and for the lethality of R. nu by inoculations at three doses. Fungal strains were applied at 1 x 10(8), 1 x 10(6), and 1 x 10(4) conidia/ml. The combinations of those insecticides with Beauveria bassiana (LPSc 1067, LPSc 1082, LPSc 1098), Metarhizium anisopliae (LPSc 907), and Metarhizium robertsii (LPSc 963) caused higher R. nu-larval mortalities than any of the individual agents alone. We observed significant differences in the in vitro conidial viability, vegetative growth, and conidia production of the five strains of entomopathogenic fungi exposed to different doses of the chemical insecticides. The combination gamma-cyhalothrin-LPSc-1067 caused the highest percent mortality of R. nu larvae, with synergism occurring between the two agents at 50% and 25% of the maximum field doses.

El uso combinado de insecticidas químicos y hongos entomopatógenos es una opción de control de plagas prometedora para minimizar los efectos químicos adversos. En este trabajo se evaluaron las interacciones entre 5 insecticidas -2 de ellos considerados biorracionales-- y 5 cepas fúngicas entomopatógenas en condiciones de laboratorio, para determinar la posible utilidad de combinaciones de estos agentes frente a R. nu. Se evaluó la compatibilidad de los insecticidas a 4 dosis mediante bioensayos in vitro y la letalidad de aquellos sobre R. nu mediante inoculaciones a 3 dosis. Las cepas fúngicas se evaluaron a concentraciones de 1 x 10(8), 1 x 10(6) y 1 x 10(4) conidios/ml. Las combinaciones de estos insecticidas con Beauveria bassiana (LPSc1067, LPSc 1082, LPSc 1098), Metarhizium anisopliae (LPSc 907) y Metarhizium robertsii (LPSc 963) causaron una mayor mortalidad de larvas de R. nu que cualquiera de los agentes individuales. Asimismo, se observaron diferencias significativas en la viabilidad de los conidios in vitro, el crecimiento vegetativo y la producción de conidios de las 5 cepas de hongos entomopatógenos expuestos a diferentes dosis de los insecticidas químicos. La combinación gamma-cialotrina-LPSc-1067 causó el mayor porcentaje de mortalidad de larvas de R. nu, con un efecto de sinergismo entre los 2 agentes al 50 y el 25% de las dosis de campo recomendadas por el fabricante.

ABSTRACT

Dual biocontrol of both insects and plant pathogens has been reported for certain fungal entomopathogens, including Beauveria bassiana and Lecanicillum spp. In this study, we demonstrate, for the first time, the dual biocontrol potential of two fungal isolates identified by morphological and phylogenetic analyses as Isaria javanica. Both these isolates caused mortality in the greater wax moth, and hence can be considered entomopathogens. Spores of the isolates were also pathogenic to nymphs of the green peach aphid (Myzus persicae), with an LC₅₀ value of 10⁷ spores/mL 4 days after inoculation and an LT₅₀ of 4.2 days with a dose of 10⁸ spores/mL. In vitro antifungal assays also demonstrated a strong inhibitory effect on the growth of two fungi that are pathogenic to peppers, Colletotrichum gloeosporioides and Phytophthora capsici. These results indicate that I. javanica isolates could be used as novel biocontrol agents for the simultaneous control of aphids and fungal diseases, such as anthracnose and Phytophthora blight, in an integrated pest management framework for red pepper.

ABSTRACT

The fruit borer Conotrachelus humeropictus is one of the most important pests of the cupuaçu crop, and can promote yield losses of around 50%. The present study involved the selection of Beauveria bassiana isolates on Conotrachelus psidii larvae, which was used as test insect due to its taxonomic closeness to C. humeropictus. In order to assess their interaction using combined application, we studied the ability of the nematode to carry conidia of B. bassiana and Metarhizium anisopliae to soil layers. This study also aimed to establish a biological control method for cupuaçu borer with the use of the fungus B. bassiana at 10 kg/ha and the entomopathogenic nematode Steinernema brazilense at 1, 3, 9, and 27 IJ/cm2, applied as single and combined treatments. In the selection of B. bassiana strains for C. psidii, the IBCB 276 and IBCB 165 isolates were the most promising ones, causing 86 and 84% mortality, respectively. The IBCB 276 strain was applied in the field to control C. humeropictus. In the study of carriage of conidia in soil, the nematode S. brazilense was found to be capable of carrying conidia of B. bassiana to deep 7 to 10 cm soil layers. This was not observed with M. anisopliae, found only in the topsoil. In field trials against C. humeropictus larvae, B. bassiana promoted 15.6% mortality. The nematode promoted 60.0% mortality at the highest concentration used, and their combined use with B. bassiana (10 kg/ha) increased mortality to 65.6%, with an additive interaction observed between the two entomopathogens.(AU)


A broca-do-cupuaçu, Conotrachelus humeropictus, é uma das principais pragas da cultura, podendo ocasionar perdas de 50% na produção. O presente trabalho envolveu uma seleção prévia de isolados de Beauveria bassiana em larvas de Conotrachelus psidii, utilizado como inseto teste por ser próximo taxonomicamente à C. humeropictus. Com o objetivo de avaliar causa-efeitos da interação na aplicação combinada, foi estudada a capacidade do nematoide carrear conídios de B. bassiana e Metarhizium anisopliae pelas camadas de solo. O trabalho também objetivou avaliar o controle biológico da broca-do-cupuaçu, através do uso do fungo B. bassiana 10 kg/ha, e do nematoide entomopatogênico Steinernema brazilense 1, 3, 9 e 27 JI/cm2, aplicados em tratamentos isolados e em combinação. Na seleção de isolados de B. bassiana para C. psidii, os isolados IBCB 276 e IBCB 165 foram os mais promissores, causando, respectivamente, 86 e 84% de mortalidade confirmada. O isolado IBCB 276 foi aplicado em campo para controlar C. humeropictus. No estudo de carreamento de conídios, o nematoide S. brazilense foi capaz de carrear conídios de B. bassiana para a camada de 7 a 10 cm de profundidade no solo. A mesma ação não foi observada com M. anisopliae, encontrado somente na camada superficial do solo. Nos ensaios de campo contra C. humeropictus, B. bassiana promoveu 15,6% de mortalidade. O nematoide promoveu 60,0% de mortalidade na maior concentração utilizada e, na combinação dessa concentração do NEP com B. bassiana (10 kg/ha), a mortalidade atingiu 65,6%, observando-se interação aditiva entre os dois entomopatógenos.(AU)

Abstract Beauveria bassiana, an entomopathogenic fungus, is the alternative biocontrol agent exploited against major economic crop pests. Pieris brassicae L. is an emerging pest of the Brassicaceae family. Therefore, in the present study, fungal isolates of Beauveria bassiana, viz. MTCC 2028, MTCC 4495, MTCC 6291, and NBAII-11, were evaluated for their virulence against third instar larvae of P. brassicae. Among all these fungal isolates, maximum mortality (86.66%) was recorded in B. bassiana MTCC 4495 at higher concentration of spores (109 conidia/ml), and the minimum mortality (30.00%) was recorded in B. bassiana MTCC 6291 at a lower concentration (107 conidia/ml) after ten days of treatment. The extracellular cuticle-degrading enzyme activities of fungal isolates were measured. Variability was observed both in the pattern of enzyme secretion and the level of enzyme activities among various fungal isolates. B. bassiana MTCC 4495 recorded the maximum mean chitinase (0.51 U/ml), protease (1.12 U/ml), and lipase activities (1.36 U/ml). The minimum mean chitinase and protease activities (0.37 and 0.91 U/ml, respectively) were recorded in B. bassiana MTCC 6291. The minimum mean lipase activity (1.04 U/ml) was recorded in B. bassiana NBAII-11. Our studies revealed B. bassiana MTCC 4495 as the most pathogenic isolate against P. brassicae, which also recorded maximum extracellular enzyme activities, suggesting the possible roles of extracellular enzymes in the pathogenicity of B. bassiana against P. brassicae.

Products were sprayed on mulberry leaves (preventive treatment) and also on the integument of silkworms (curative treatment). At the first stage, the products were applied on four consecutive days, in one of the daily feeds. Planta Clean® - curative treatment - and Soil-Set® - preventive treatment - caused mortality in 63.3% and 30% of silk worms, respectively. The fungus Beauveria bassiana was inoculated at the concentration 5 × 107 conidia/mL on the leaves and insects. The products were applied three hours after application on the fungus, in a single application, and also for four days in the other groups. A group of silkworms sprayed only with the fungus served as control. Biogermex® - curative treatment applied once and for four times - was the most effective, reducing mortality by 25 and 26.7%, respectively, as compared with the control group. None of the evaluated products acts preventively against white muscardine.


Este trabalho objetivou avaliar os efeitos de produtos Biogermex®, Agro-mos®, Soil-Set® e Planta Clean® no desenvolvimento da calcinose branca em Bombyx mori. Os produtos foram testados em duas etapas: efeitos sobre a biologia e produção de seda pela lagarta (1) e, em seguida, em função do seu potencial na prevenção/no controle da calcinose branca (2). Os produtos foram pulverizados sobre as folhas de amoreira (tratamento preventivo) e sobre o tegumento das lagartas (tratamento curativo). Na primeira etapa, os produtos foram aplicados durante quatro dias consecutivos, em uma das alimentações do dia. Planta Clean® - tratamento curativo - e Soil-Set® - tratamento preventivo - causaram mortalidade em 63,3 e 30% das lagartas, respectivamente. ABSTRACT

Beauvericin (BEA), a cyclic hexadepsipeptide produced by the fungus Beauveria bassiana, is known to have anti-cancer, anti-inflammatory, and anti-microbial actions. However, how BEA suppresses macrophage-induced inflammatory responses has not been fully elucidated. In this study, we explored the anti-inflammatory properties of BEA and the underlying molecular mechanisms using lipopolysaccharide (LPS)-treated macrophage-like RAW264.7 cells. Levels of nitric oxide (NO), mRNA levels of transcription factors and the inflammatory genes inducible NO synthase (iNOS) and interleukin (IL)-1, and protein levels of activated intracellular signaling molecules were determined by Griess assay, semi-quantitative reverse transcriptase-polymerase chain reaction (RT-PCR), luciferase reporter gene assay, and immunoblotting analysis. BEA dose-dependently blocked the production of NO in LPS-treated RAW264.7 cells without inducing cell cytotoxicity. BEA also prevented LPS-triggered morphological changes. This compound significantly inhibited nuclear translocation of the NF-κB subunits p65 and p50. Luciferase reporter gene assays demonstrated that BEA suppresses MyD88-dependent NF-κB activation. By analyzing upstream signaling events for NF-κB activation and overexpressing Src and Syk, these two enzymes were revealed to be targets of BEA. Together, these results suggest that BEA suppresses NF-κB-dependent inflammatory responses by suppressing both Src and Syk.

ABSTRACT

The green peach aphid (Myzus persicae), a plant pest, and gray mold disease, caused by Botrytis cinerea, affect vegetables and fruit crops all over the world. To control this aphid and mold, farmers typically rely on the use of chemical insecticides or fungicides. However, intensive use of these chemicals over many years has led to the development of resistance. To overcome this problem, there is a need to develop alternative control methods to suppress populations of this plant pest and pathogen. Recently, potential roles have been demonstrated for entomopathogenic fungi in endophytism, phytopathogen antagonism, plant growth promotion, and rhizosphere colonization. Here, the antifungal activities of selected fungi with high virulence against green peach aphids were tested to explore their potential for the dual control of B. cinerea and M. persicae. Antifungal activities against B. cinerea were evaluated by dual culture assays using both aerial conidia and cultural filtrates of entomopathogenic fungi. Two fungal isolates, Beauveria bassiana SD15 and Metarhizium anisopliae SD3, were identified as having both virulence against aphids and antifungal activity. The virulence of these isolates against aphids was further tested using cultural filtrates, blastospores, and aerial conidia. The most virulence was observed in the simultaneous treatment with blastospores and cultural filtrate. These results suggest that the two fungal isolates selected in this study could be used effectively for the dual control of green peach aphids and gray mold for crop protection.

Abstract This study was performed under laboratory conditions to identify isolates of the fungus Beauveria spp. that can control Polyphagotarsonemus latus in the greenhouse and field. Thirty Beauveria spp. isolates were tested by spraying 1 mL conidia (1 × 108 conidia/mL) on pepper leaf discs containing 15 mites. Evaluations were performed on the 3rd and 6th day post application by counting the number of dead mites. Vegetative growth and conidial production were measured from the selected isolates, and bioassays were conducted in the greenhouse on bean seedlings in plastic pots. The isolate Unioeste 53 was selected, and a conidial suspension (1 × 108 conidia/mL) was applied with a backpack sprayer. The evaluation consisted of pre- and post-treatment counts of the number of live mites on ten leaflets in both the plots treated with the fungus and control plots, and the same procedure was followed for the field experiment. In the laboratory, the Unioeste 53 isolate resulted in total and confirmed mortality rates of 70% and 57.7%, respectively. In the greenhouse, the population decreased by 76.71% by the 16th day after application. In the field, the population decreased by 66% by the 12th day after application, demonstrating the potential of this fungus for mite management.


Resumo Este trabalho teve por objetivo selecionar isolados do fungo Beauveria bassiana em condições de laboratório com potencial de uso em casa de vegetação e campo no controle do ácaro branco. Foi realizada uma seleção com 30 isolados de Beauveria spp. através de pulverização direta de conídios (1 × 108 conídios/mL) sobre discos foliares de pimenta contendo 15 ácaros. As avaliações foram realizadas no terceiro e sexto dia contando-se o número de mortos, e confirmação do patógeno em câmara úmida. Parâmetros de crescimento vegetativo e produção de conídios foram avaliados. No bioensaio em casa de vegetação foram preparados vasos com plantas de feijão e fez-se a aplicação do Unioeste 53 (1 × 108 conídios/mL), utilizando pulverizador costal, a avaliação constou da contagem prévia e posterior à aplicação do número de ácaros vivos em 10 folíolos, tanto nas parcelas destinadas ao tratamento com fungo, quanto na testemunha. A fase de campo seguiu os mesmos padrões, porém, com área experimental total de 225 m2, com oito parcelas de 10,24 m2, sendo 4 testemunhas e 4 onde foi aplicado o isolado Unioeste 53 seguindo metodologia de aplicação e avaliação já descritas para casa de vegetação. Em laboratório o isolado Unioeste 53 causou mortalidade total de 70% e 57,7% de mortalidade confirmada. Em casa de vegetação, apresentou redução da população de 76,71% 16 dias após aplicação, já em campo, a redução da população foi de 66% após 12 dias da aplicação, demonstrando o potencial do ácaro pelo fungo.
